The first article was about the discovery of a new transcriptional co-activator. During a screening of cDNA for molecules that bind 14-3-3, Kanai et al. (2000) discovered the molecule TAZ. TAZ stands for transcriptional co-activator with PDZ-binding motif. Through analyzing TAZ they were able to identify its binding mechanism to 14-3-3. It was discovered that the PDZ motif on TAZ binds to the protein NHERF-2, and overall can be a part of nuclear transcription at the plasma membrane and cytoskeleton. From there, Kanai et al. determined that 14-3-3 could be a potential regulatory tool for TAZ at the sites.

The article “TAZ: a novel transcriptional co-activator regulated by interactions with 14-3-3 and PDZ domain proteins” (Kanai et al., 2000) introduces a new molecule into the scientific world. Co-activators and regulatory proteins are concepts that I am familiar with from Biochemistry 3400. We learn that co-activators bind to activators to increase the rate of transcription and that regulatory proteins help control the synthesis of proteins. I think it’s really cool that through a simple screening procedure Kanai et al. were able to discover a molecule that may be heavily involved in the transcription of genes at sites other than the plasma membrane and cytoskeleton whilst finding a potential regulatory protein for these reactions. As it pertains to Dr. Marignani’s research, TAZ may not be a tumour suppressor protein but its discovery produced a molecule that plays a role in the human body. Through more tests and analysis of TAZ, there may be a link between TAZ, tumor suppressor proteins and cellular equilibrium. TAZ also has a connection to events regarding nuclear transcription at the cytoskeleton. Cytoskeletal reorganization is something that Dr. Marignani researches, so TAZ may become a molecule worth more research.
